Depay, a January signing from Manchester United, hit Lyon's third and fourth goals in the Ligue 1 win, with the latter coming after he picked up the ball just inside Toulouse's half before looping a brilliant effort into the air and over the stranded Alban Lafont.
Christophe Jallet had fired Lyon into the lead in the 36th minute from a tight angle, before Maxwel Cornet made it two with a strike into the top corner just after the break.
Depay got in on the act after 53 minutes after being played in by Jallet, with the Netherlands international then scoring his stunning second eight minutes from the end.
Loic Perrin rescued a point for St Etienne with his last-minute strike securing a 2-2 draw against Metz.
Ismaila Sarr had opened the scoring for Metz inside the first minute with a hit from distance, only for Robert Beric to equalise for the hosts eight minutes after the break.
Metz looked to be heading for victory following Simon Falette's 67th-minute close-range finish, but Perrin popped up to head home in the 95th minute to salvage a share of the spoils for St Etienne.
